A new series of Schiff bases derived from pyrazole-thiocarbohydrazide, namely (4a-d), were well-synthesized. The synthesis is carried out using monothiocarbohydrazone derivative (3), which was prepared via coupling of 5-chloro-pyrazole-4-carbaldehyde (1) with thiocarbohydrazide (2) in absolute ethanol that contains a catalytic quantity of acetic acid. The structures of newly synthesized compounds were fully clarified by various spectroscopic analyses (FT-IR, ¹H-NMR, ¹³C-NMR, and mass spectra) and elemental analysis. Also, the molecular docking was performed to investigate the binding interactions of the synthesized compounds (1, 3 and 4a-d) with COX-2 active site. The results revealed that most of them have robust hydrogen bonding networks and favorable binding energies compared to compound (4b). Understanding the anti-inflammatory behavior through understanding the specific interactions of these compounds with COX-2 will aid in the design and development of more effective inhibitors for therapeutic applications.

The present paper proposed the tri-potassium phosphate catalyzed synthesis of some novel series of Schiff bases (3a to 3l) derived from 2-aminopyridine using microwave technique. All synthesized Schiff bases were confirmed by different spectroscopic methods. Further all synthesized imines have been evaluated to screen for antimicrobial, antioxidant and anti-inflammatory activity. The compound 3c displays potent anti-inflammatory activity with 42.307%±1.322 inhibition of inflammation. The antioxidant activity data revealed that all synthesized Schiff bases exhibit potency to scavenge DPPH and nitric oxide radicals. Antimicrobial properties of these compounds showed moderate to excellent growth of inhibitory activity against various tested pathogens. Our observations and examination for pharmacological study towards the presence of active pharmacological functional groups present in the main structural nucleus of synthesized imines. Therefore, present study may lead to the development of a new class of anti-inflammatory and antioxidant drugs with structural modification.

Here derivatives of imidazo[2,1-b][1,3]thiazines are attractive objects for organic and medicinal chemists. In the present work chemoselective conditions for oxidation of the sulfur atom in the 6-(2-pyridinyloxy)substituted (benzo)imidazo[2,1-b][1,3]thiazines to the corresponding sulfoxides were proposed and their synthesis was performed. Synthesized sulfoxides exist in the diastereomeric mixture and individual diastereomers 2a-e and 3a-e were obtained using a chromatographic technique. The structure of compounds 2a-e and 3a-e were characterized using ¹H, ¹³C NMR, LC-MS spectra, and X-ray analysis for derivative 2b. The anti-inflammatory activity screening in vivo was performed using the carrageenan model of inflammatory paw edema in white rats for all the diastereomeric mixtures and individual diastereomers. Diastereomer 2c possessed an anti-inflammatory effect with an inflammation inhibition index of 46.1% which was equal to the activity of the reference drug diclofenac sodium.
